Drug intelligence / Profile preview
Irinotecan liposomal (also known as nanoliposomal irinotecan or nal-IRI) is a nanotherapeutic formulation of the topoisomerase I inhibitor irinotecan. The drug is encapsulated in long-circulating liposome-based nanoparticles using a proprietary intraliposomal stabilization technology, which allows for high drug loading and sustained release. This formulation is designed to improve the pharmacokinetics and biodistribution of irinotecan, protecting it from premature metabolism into its active metabolite, SN-38, and enhancing its accumulation within tumor tissues. It was originally developed by Merrimack Pharmaceuticals as MM-398 and is currently marketed by Ipsen. It is FDA-approved for the treatment of metastatic pancreatic adenocarcinoma in combination with fluorouracil and leucovorin following progression on gemcitabine-based therapy.
