Irinotecan liposome II is a liposomal formulation of the topoisomerase I inhibitor irinotecan, developed by CSPC Pharmaceutical Group (specifically its subsidiary CSPC Ouyi Pharmaceutical). It was approved by China's National Medical Products Administration (NMPA) in September 2023 under the brand name Duoenda. The (II) designation distinguishes this specific formulation from other liposomal irinotecan products. It is indicated for use in combination with 5-fluorouracil and leucovorin for the treatment of patients with metastatic pancreatic adenocarcinoma who have progressed following gemcitabine-based therapy. The liposomal delivery system is designed to enhance the drug's stability in circulation and increase its accumulation in tumor tissues, where the prodrug irinotecan is converted into the active metabolite SN-38. SN-38 then inhibits DNA topoisomerase I, leading to lethal DNA strand breaks and tumor cell apoptosis.
