Overview
Irinotecan liposome injection is a nanoliposomal formulation of the topoisomerase I inhibitor irinotecan, designed for intravenous administration. Encapsulation in a lipid bilayer (liposome) prolongs intratumoral exposure to the active metabolite SN-38, enhancing antitumor activity and potentially reducing systemic toxicity compared to conventional irinotecan. The drug acts by inhibiting topoisomerase I, an enzyme essential for DNA replication; this leads to DNA damage and cell death in rapidly dividing cancer cells. Irinotecan liposome injection is primarily indicated, in combination with fluorouracil and leucovorin, for adults with metastatic pancreatic adenocarcinoma that has progressed following gemcitabine-based therapy. It has also been studied in other cancers such as colorectal cancer and gastric cancer[1][2][3][4][5][6].
