Drug intelligence / Profile preview
Irinotecan pegylated liposomal is a **pegylated liposomal formulation of irinotecan** designed for intravenous administration as an antineoplastic agent. Irinotecan is a prodrug that is metabolized to SN-38, its active form, which inhibits **topoisomerase 1**—an enzyme essential for DNA replication and transcription. Encapsulation in PEGylated liposomes enhances the plasma half-life of irinotecan, stabilizes the drug in its active lactone form, and enables more selective accumulation in tumor tissues through the enhanced permeability and retention effect, improving therapeutic efficacy and reducing systemic toxicity compared to conventional irinotecan. Developed as a treatment for metastatic pancreatic cancer, especially after progression on gemcitabine-based therapies, it is also approved for first-line use in combination regimens including oxaliplatin, fluorouracil, and leucovorin. The primary developer and manufacturer is Ipsen Biopharmaceuticals. The drug is not interchangeable with regular (non-liposomal) irinotecan[1][3][7][9][12].
