Drug intelligence / Profile preview
Irodanoprost (MES1022) is a bone-targeted prodrug of a selective prostaglandin E2 receptor 4 (EP4) agonist developed by Mesentech. It consists of a potent EP4-selective agonist (MES1002) reversibly conjugated via a hydrolyzable ester linker to a bisphosphonate-based bone-binding moiety. This tissue-targeting design allows the drug to selectively distribute to calcium-rich tissues, such as bone and damaged myofibers, minimizing systemic exposure and off-target side effects. Upon esterase-mediated hydrolysis, the active EP4 agonist moiety is released. Irodanoprost is being developed for the treatment of bone fractures, osteoporosis, osteogenesis imperfecta, and muscular dystrophies, including Duchenne muscular dystrophy (DMD) and facioscapulohumeral muscular dystrophy (FSHD).
