Overview
Irofulven (LP-100) is a semisynthetic derivative of illudin S, a natural toxin isolated from the mushroom *Omphalotus illudens* (Jack O'Lantern mushroom). It functions as a DNA-damaging agent that covalently binds to DNA, forming bulky adducts that interfere with DNA replication and transcription. These adducts are primarily repaired by the nucleotide excision repair (NER) pathway; consequently, irofulven exhibits enhanced cytotoxicity in tumor cells with deficient NER mechanisms. Originally developed by MGI Pharma and later licensed to Lantern Pharma, the compound has been evaluated in numerous clinical trials for solid tumors and leukemias. In 2021, Lantern Pharma reacquired global rights from Allarity Therapeutics to advance the drug using its RADR® AI platform, which leverages biomarker-driven development to identify patients most likely to respond, particularly in metastatic castration-resistant prostate cancer (mCRPC) and bladder cancer.
