IONP-GA/PAA is an iron-based polymeric nano-formulation consisting of an iron oxide nanoparticle core coated with gallic acid (GA) and polyacrylic acid (PAA). It is designed to treat various cancers by inducing ferroptosis, a form of regulated cell death characterized by iron-dependent lipid peroxidation. The formulation has demonstrated intrinsic cytotoxic activity in preclinical models of glioblastoma, neuroblastoma, and fibrosarcoma. Its mechanism involves the promotion of lipid hydroperoxide generation, which leads to oxidative stress and subsequent cell death. This activity can be specifically inhibited by iron chelators and lipophilic radical traps, confirming the specificity of the ferroptotic pathway induction. The nanoparticle is currently in the preclinical stage of development, with potential for further in vivo validation.
