Irradiated 4T1.2 whole tumor cell vaccine is an experimental, preclinical cancer vaccine consisting of lethally irradiated 4T1.2 mouse mammary tumor cells. The 4T1.2 cell line is a highly metastatic, poorly immunogenic subclone of the 4T1 murine breast cancer model, which closely mimics human stage IV triple-negative breast cancer. By irradiating the cells, their proliferative capacity is destroyed while preserving their cell-surface tumor-associated antigens. When administered as a vaccine, it is designed to stimulate the host immune system, promoting tumor-specific interferon-gamma (IFN-γ) production, enhancing CD4+ and CD8+ T-cell responses, and reducing the accumulation of immunosuppressive splenic myeloid-derived suppressor cells (MDSCs). It has been studied preclinically, often in combination with lifestyle interventions (such as diet and exercise-induced weight maintenance) or immune checkpoint inhibitors (like anti-PD-1 antibodies), to delay primary mammary tumor growth and reduce metastatic burden.
