Irradiated allogeneic lymphocytes is an investigational cell-based immunotherapy developed by Rutgers, The State University of New Jersey, for the treatment of metastatic renal cell carcinoma. The therapy consists of partially HLA-matched blood mononuclear cells (lymphocytes) obtained from relatives of the patient. These cells are irradiated prior to infusion to prevent graft-versus-host disease (GVHD) while retaining their ability to induce an allogeneic immune response against the tumor. This mechanism mimics the graft-versus-tumor (GVT) effect observed in hematopoietic stem cell transplantation but is administered as a standalone biologic therapy. It has been evaluated in Phase II clinical trials (such as the SPECIAL trial) in combination with the tyrosine kinase inhibitor sunitinib for patients with metastatic clear cell renal cell carcinoma.
