Irradiated donor-derived CD19-zeta-transduced EBV-lymphoblastoid cell line is a cellular immunotherapy developed by University College London for the treatment of high-risk or relapsed B-cell acute lymphoblastic leukemia (ALL) following allogeneic hematopoietic stem cell transplantation. The product consists of lymphoblastoid cell lines (LCLs) generated from the stem cell donor by infecting B-cells with Epstein-Barr Virus (EBV). These LCLs are genetically modified with a retroviral vector (SFGalpha-CD19-CD3zeta) to express a CD19-specific chimeric antigen receptor (CAR) and are subsequently irradiated (typically at 70Gy) to render them non-proliferative while maintaining their antigenicity. Administered subcutaneously, these cells function as a cellular vaccine designed to provide antigenic stimulation to infused CD19-zeta gene-modified EBV-specific cytotoxic T-lymphocytes (CTLs), thereby promoting their expansion and long-term persistence in vivo.
