IRS 954 is a synthetic immunoregulatory oligodeoxynucleotide (INH-ODN) that functions as a dual antagonist of Toll-like receptor 7 (TLR7) and Toll-like receptor 9 (TLR9). Developed by Dynavax Technologies, it is designed to block the activation of the innate immune system by self-nucleic acids, a key driver in the pathogenesis of systemic lupus erythematosus (SLE) and lupus nephritis. By inhibiting TLR7 and TLR9 signaling in B cells and plasmacytoid dendritic cells, IRS 954 reduces the production of type I interferons and autoantibodies. Preclinical studies in lupus-prone mouse models, such as MRL/lpr mice, have shown that IRS 954 can prevent the progression of renal disease, attenuate lung injury, and improve overall survival. While it served as a significant lead compound, it has primarily been utilized in experimental research to explore the role of TLR signaling in autoimmunity.
