Overview

IRX4204 is an orally bioavailable, highly selective, and potent second-generation agonist of the retinoid X receptor (RXR), also known as a rexinoid. It activates RXR homodimers and RXR/Nurr1 heterodimers at nanomolar concentrations without significant activity on other nuclear receptors. The drug exhibits antineoplastic, neuroprotective, immunoregulatory, and disease-modifying activities. Preclinical studies have shown that IRX4204 promotes survival of dopaminergic neurons and supports myelin repair. Clinically, IRX4204 has been evaluated in phase I/II trials for various cancers and Parkinson’s disease and has demonstrated safety with long-term oral dosing. It is being developed by Io Therapeutics. Ongoing development includes planned phase II trials for amyotrophic lateral sclerosis (ALS) and consideration for Alzheimer’s disease.
