IRX4647 is a selective small molecule agonist of the retinoic acid receptor-gamma (RARγ) currently in preclinical development. As a synthetic retinoid derivative, it is being investigated as a novel cancer immunotherapy agent, particularly for its potential to overcome resistance to immune checkpoint inhibitors. Preclinical studies in syngeneic lung cancer models have demonstrated that IRX4647, when used in combination with anti-PD-L1 therapy, can significantly suppress tumor growth compared to checkpoint inhibition alone. Beyond its immunomodulatory properties, IRX4647 has shown the ability to inhibit RANKL-stimulated osteoclastogenesis, suggesting potential applications in treating bone-related pathologies or bone metastases. The compound is characterized by a plasma half-life of approximately 6 hours in murine models and requires further toxicology and pharmacokinetic evaluation before advancing to clinical trials.
