Overview

IS154 is a second-generation imidazolium compound being developed as a tumor-specific intravesical therapeutic agent for the treatment of high-risk non-muscle invasive bladder cancer (NMIBC). Developed by researchers at Fox Chase Cancer Center and the University of Akron, IS154 is designed to address the limitations of current intravesical therapies like BCG, which face frequent shortages. The compound exhibits selective toxicity toward tumor tissues while sparing normal urothelium. Its mechanism of action involves the induction of apoptosis through the mitochondrial pathway. In preclinical studies, IS154 demonstrated high potency against various bladder cancer cell lines and organoids without causing histopathological changes in murine bladder urothelium after intravesical instillation.
