Isaria sinclairii ethanol extract is a natural product derived from the entomopathogenic fungus Isaria sinclairii (also known as Cordyceps sinclairii). It is historically significant as the source of myriocin (ISP-I), the structural lead compound for the development of the sphingosine-1-phosphate receptor modulator fingolimod (FTY720). The extract exhibits potent immunosuppressive and anti-inflammatory activities, primarily through the inhibition of serine palmitoyltransferase, the rate-limiting enzyme in sphingolipid biosynthesis. This inhibition leads to a reduction in sphingosine-1-phosphate levels, which affects lymphocyte trafficking. Research has also explored the extract's potential in treating metabolic disorders, such as obesity and type 2 diabetes, by modulating lipid metabolism and suppressing pro-inflammatory cytokines in adipose tissue.
