Overview

Isatuximab + carfilzomib + dexamethasone is a three-drug combination regimen used primarily for the treatment of relapsed or refractory multiple myeloma. Isatuximab is a monoclonal antibody targeting CD38 on plasma cells, leading to cell death through immune-mediated mechanisms and direct apoptosis. Carfilzomib is a selective proteasome inhibitor that disrupts protein degradation in cancer cells, resulting in apoptosis. Dexamethasone is a synthetic glucocorticoid with anti-inflammatory and immunosuppressive properties that also induces apoptosis in certain hematologic malignancies. This combination has demonstrated significant efficacy in prolonging progression-free survival for patients who have received one to three prior lines of therapy[2][6][8]. The regimen is administered intravenously on a 28-day cycle until disease progression or unacceptable toxicity.
