Isatuximab + lenalidomide is a combination therapy primarily used for the treatment of multiple myeloma, particularly in patients who are newly diagnosed and not eligible for autologous stem cell transplant, as well as in relapsed or refractory cases. Isatuximab is a monoclonal antibody that targets CD38, a glycoprotein highly expressed on malignant plasma cells, leading to their apoptosis through mechanisms such as antibody-dependent cellular cytotoxicity (ADCC) and complement-dependent cytotoxicity. Lenalidomide is an immunomodulatory small molecule that alters cytokine production, enhances T-cell and natural killer (NK) cell activity, inhibits angiogenesis, and induces degradation of key transcription factors essential for myeloma cell survival by modulating the cereblon E3 ubiquitin ligase complex. The combination has demonstrated efficacy in clinical trials for multiple myeloma[1][2][3][5][6][8].
