isavuconazole + methotrexate

**isavuconazole + methotrexate** is a combination of two drugs: isavuconazole, a broad-spectrum triazole antifungal agent, and methotrexate, an antimetabolite and immunosuppressant often used in chemotherapy and autoimmune diseases. Isavuconazole inhibits cytochrome P450-dependent lanosterol 14-alpha-demethylase, thereby blocking the synthesis of ergosterol, a key component of fungal cell membranes, leading to cell death. Methotrexate is a substrate of breast cancer resistance protein (BCRP), organic anion transporter 1/3 (OAT1/OAT3), and acts by inhibiting dihydrofolate reductase, interfering with DNA synthesis, repair, and cellular replication.
