ISC-4 is a synthetic small-molecule phenylbutyl isoselenocyanate designed as a selenium-containing analog of naturally occurring phenylalkyl isothiocyanates to potently inhibit the PI3K/Akt signaling pathway in cancer cells. It directly suppresses phosphorylated Akt, activates the tumor suppressor Par-4, induces mitochondrial-dependent, caspase-9/3–mediated apoptosis, and selectively kills malignant cells while sparing normal cells in preclinical models of colon cancer, melanoma, and acute myeloid leukemia. ISC-4 has also been shown to modulate xenobiotic metabolism by inhibiting cytochrome P450 enzymes and inducing phase II detoxification enzymes, and demonstrates chemopreventive and therapeutic activity in multiple murine tumor models with limited observed systemic toxicity.[1][3][5][7][13][15]
