Overview

This is a **combination therapy** comprising iscalimab, mycophenolate mofetil, and corticosteroids. - **Iscalimab** is a fully human monoclonal antibody targeting CD40, a costimulatory protein on antigen-presenting cells (APCs) that is important for immune cell activation. By blocking CD40 and its ligand CD40L (CD154), iscalimab interrupts immune cell activation, inhibits production of pro-inflammatory cytokines (e.g., IL-6, TNF-α, IFN-γ), curtails germinal center formation, and reduces autoantibody production. It is being developed primarily as an immunosuppressive agent for transplant rejection and autoimmune diseases[2][5][8]. - **Mycophenolate mofetil** is an immunosuppressant that inhibits inosine monophosphate dehydrogenase (IMPDH), thereby preventing lymphocyte proliferation. It is widely used to prevent organ rejection and in various autoimmune indications[4]. - **Corticosteroids** are synthetic or natural steroid hormones that are anti-inflammatory and immunosuppressive. They inhibit transcription of genes encoding pro-inflammatory cytokines and other molecules, acting via the glucocorticoid receptor[3][6]. This combination is being explored as treatment for severe autoimmune diseases and for transplantation, notably including thyroid eye disease and kidney transplant settings[1][7].
