Overview

ISCOMATRIX is a novel vaccine adjuvant composed of saponin (derived from Quillaja saponaria), cholesterol, and phospholipid arranged in a particulate structure. Unlike traditional immunostimulatory complexes (ISCOMs), it does not contain an incorporated antigen but serves as a delivery system and immune enhancer when mixed with antigens. Its mechanism involves rapid trafficking of antigen into the cytosol of dendritic cells, induction of cytokines and chemokines, and linking innate to adaptive immunity in a Toll-like receptor-independent but MyD88-dependent manner. It induces broad humoral (antibody) and cellular (CD4+ and CD8+ T cell) responses—including strong cytotoxic T lymphocyte activity—and can be used for both prophylactic and therapeutic vaccines against infectious diseases or cancer. Clinical trials have shown it to be generally safe, well tolerated, and immunogenic[1][2][3][5][7][8].
