Overview

ISIKOK-19 is an academic chimeric antigen receptor (CAR) T-cell therapy developed in Turkey for the treatment of CD19-positive B-cell malignancies. It is a second-generation CAR-T product utilizing a lentiviral vector to express a CAR construct consisting of an FMC63-derived CD19-specific single-chain variable fragment (scFv), a CD8-derived hinge and transmembrane domain, a CD28 costimulatory domain, and a CD3ζ signaling domain. The construct also includes a truncated human epidermal growth factor receptor (EGFRt) which serves as a selection marker and a potential safety switch. The manufacturing process is notable for the use of BX-795, a TBK1/IKKƐ inhibitor, to enhance lentiviral transduction efficiency and reduce production costs, facilitating its use as a more affordable academic alternative to commercial CAR-T products.
