ISIS 141923 is a synthetic antisense oligonucleotide (ASO) designed as a negative control for preclinical and pharmacological studies. It is a 20-nucleotide phosphorothioate oligonucleotide with full 2'-O-(2-methoxyethyl) (2'-MOE) modifications at the ends and deoxynucleosides in the central region. The sequence of ISIS 141923 does not have perfect complementarity to any known gene in public databases and thus does not target or modulate any specific mRNA. Its primary use is as a "scrambled" or non-targeting control in experiments evaluating the effects of other gene-specific antisense oligonucleotides. Developed by Ionis Pharmaceuticals (formerly Isis Pharmaceuticals), it has been widely used to demonstrate specificity and safety profiles of therapeutic ASOs[1][2][6][7].
