Overview

ISIS 183750 + irinotecan is a combination therapy evaluated primarily for advanced solid tumors and refractory colorectal cancer. ISIS 183750 is a second-generation antisense oligonucleotide designed to inhibit the production of eukaryotic translation initiation factor 4E (eIF4E), an oncogene dysregulated in many cancers. By reducing eIF4E mRNA and protein levels, it aims to suppress tumor cell proliferation. Irinotecan is a topoisomerase I inhibitor chemotherapy agent that induces DNA damage in rapidly dividing cells, leading to cell death. Preclinical studies demonstrated additive antiproliferative effects when these agents are combined. Clinical trials have shown that this combination can achieve disease stabilization in patients with refractory colorectal cancer who had progressed on prior therapies[1][3][2].
