ISIS-3082 is a murine-specific phosphorothioate antisense oligonucleotide (ASO) designed to inhibit the expression of Intercellular Adhesion Molecule 1 (ICAM-1). Developed by Ionis Pharmaceuticals (formerly Isis Pharmaceuticals), it serves as the mouse surrogate for the human ICAM-1 inhibitor alicaforsen (ISIS 2302). ISIS-3082 works by binding to the ICAM-1 mRNA transcript, promoting its degradation via RNase H and thereby reducing the cell-surface expression of ICAM-1 protein. This molecule is widely used in preclinical research to study the role of ICAM-1 in inflammatory diseases, organ transplant rejection, and more recently, the modulation of the tumor microenvironment in glioblastoma. By inhibiting ICAM-1, it can prevent the infiltration of certain immune cell subsets and alter the polarization of tumor-associated macrophages towards an M1-like state.
