Overview

ISIS-329993 is a **second-generation antisense oligonucleotide** developed to inhibit the production of *C-reactive protein* (CRP) by binding specifically to CRP mRNA and preventing its translation. CRP is a marker and mediator of inflammation implicated in several diseases, including cardiovascular and autoimmune conditions. ISIS-329993 was primarily investigated in phase 2 clinical trials for its ability to lower CRP levels in patients with paroxysmal atrial fibrillation and rheumatoid arthritis. While ISIS-329993 led to dose-dependent reductions in CRP, trials did not show an improvement in disease activity or symptom burden in these settings. The drug demonstrated favorable safety and tolerability profiles but was ultimately **discontinued in clinical development**[1][2][3][7].
