ISIS 353512 is an antisense oligonucleotide drug developed to inhibit the expression of C-reactive protein (CRP), a biomarker and mediator of inflammation. The drug was designed to bind specifically to the messenger RNA (mRNA) encoding human CRP, thereby reducing its production. It was primarily investigated for potential therapeutic benefit in cardiovascular, renal, and inflammatory diseases by lowering CRP levels[1][2][3]. Clinical development included phase I trials in healthy volunteers; however, unexpected immune activation—specifically increases in interleukin-6 (IL-6) and CRP—was observed due to B cell activation likely mediated by Toll-like receptor 9 (TLR9)[2][6][8]. These immunostimulatory effects led to discontinuation of its development.
