Overview

This is a combination regimen consisting of three agents: **ISIS 5132** is a synthetic, 20-base phosphorothioate antisense oligonucleotide designed to bind specifically to c-raf mRNA, thereby inhibiting the expression of Raf-1 serine/threonine kinase. Raf kinases are key effectors in the Ras/mitogen activated protein kinase pathway and play an essential role in cell proliferation and oncogenic transformation. By downregulating c-Raf expression, ISIS 5132 aims to suppress tumor growth[1][3][5]. **Carboplatin** is a platinum-based chemotherapeutic agent that causes DNA crosslinking and apoptosis in rapidly dividing cells[6][8]. **Paclitaxel** is a microtubule-stabilizing agent that inhibits cell division by preventing microtubule depolymerization during mitosis[6][8]. The combination has been investigated primarily for advanced or recurrent solid tumors such as ovarian cancer. The rationale for combining these drugs is to target cancer cells through multiple mechanisms—gene expression inhibition (ISIS 5132), DNA damage (carboplatin), and mitotic arrest (paclitaxel)[1][5].
