Overview

ISM8969 is an orally available, brain-penetrant small molecule inhibitor of the NLRP3 (NOD-like receptor protein 3) inflammasome, discovered using Insilico Medicine's generative AI platform, Chemistry42. It is specifically designed to cross the blood-brain barrier to target neuroinflammation, which is a key driver of neurodegenerative conditions such as Parkinson's disease and Alzheimer's disease. In January 2026, Insilico Medicine entered into a global co-development and licensing agreement with Hygtia Therapeutics (a Fosun Pharma incubatee) to advance the drug. Under the terms of the agreement, Insilico is responsible for the Investigational New Drug (IND) submission and Phase 1 clinical trials for Parkinson's disease, while Hygtia will lead subsequent clinical development, regulatory filings, and commercialization. Preclinical data have demonstrated that ISM8969 provides dose-dependent improvements in motor function in Parkinson's models and exhibits a favorable safety and pharmacokinetic profile.
