Overview

Isoaminile is a synthetic small molecule antitussive (cough suppressant) structurally related to methadone. It acts centrally to suppress the cough reflex by influencing the cough center in the central nervous system. Isoaminile also exhibits anticholinergic activity, functioning as both an antimuscarinic and antinicotinic agent, which contributes to its therapeutic effects by inhibiting acetylcholine at muscarinic receptors and reducing bronchial secretions. It is typically administered as a cyclamate or citrate salt, with common brand names including Peracon, Aprecon, and Nullatuss. The drug has been approved for use in treating cough but has also been withdrawn in some regions[1][2][3][5][6][7][8].
