Overview

Isobutyl-deoxynyboquinone (IB-DNQ) is a small molecule quinone derivative that functions as a potent substrate for NAD(P)H-quinone oxidoreductase 1 (NQO1). In NQO1-overexpressing cells, such as those found in fumarate hydratase (FH)-deficient renal cell carcinoma, IB-DNQ undergoes a futile redox cycle. This cycle consumes oxygen and generates high levels of reactive oxygen species, specifically superoxide anions, while rapidly depleting cellular pools of NADH and NADPH. The resulting metabolic stress suppresses aerobic glycolysis and reduces ATP levels, leading to tumor growth inhibition. IB-DNQ is being investigated for the treatment of hereditary leiomyomatosis and renal cell carcinoma (HLRCC), where NQO1 is upregulated due to NRF2 activation following fumarate accumulation.
