Drug intelligence / Profile preview
**Isomesothelin-CAR-iNKT** is an investigational **allogeneic chimeric antigen receptor invariant natural killer T cell therapy** engineered to recognize **Isomesothelin**, an alternatively spliced mesothelin variant expressed on malignant cells. In published preclinical conference data, expanded and genetically modified iNKT cells targeting IsoMSLN showed cytotoxic activity and a central-memory-like phenotype, supporting the concept of an off-the-shelf cell therapy platform for solid tumors. The therapeutic mechanism is expected to combine CAR-mediated recognition of the Isomesothelin antigen with the intrinsic antitumor biology of iNKT cells, which can recognize lipid antigens presented by CD1d and may have lower graft-versus-host disease risk than conventional allogeneic T-cell products. Available evidence appears to be preclinical, and the program has been associated with Kiromic's broader IsoMesothelin cell-therapy efforts, although direct clinical-stage evidence specific to the iNKT version is limited.
