Isometheptene is a sympathomimetic amine used primarily for the treatment of migraines and tension headaches due to its vasoconstricting properties. It acts by constricting dilated cranial and cerebral arterioles, thereby reducing the stimuli that lead to vascular headaches. Isometheptene exerts its effects through activation of the sympathetic nervous system via epinephrine and norepinephrine, which interact with adrenergic receptors on smooth muscle cells, leading to increased intracellular calcium and subsequent vasoconstriction. The drug may also have a mixed mechanism involving both tyramine-like action (indirectly releasing catecholamines) and direct stimulation of α1-adrenoceptors[1][5][6]. Isometheptene is often formulated as the mucate salt in combination products for headache relief.
