Isophane human insulin, also known as NPH (Neutral Protamine Hagedorn) insulin, is an intermediate-acting human insulin biologic developed by Novo Nordisk for the treatment of diabetes mellitus. It is produced using recombinant DNA technology and is structurally identical to endogenous human insulin. The formulation consists of a cloudy suspension of insulin with protamine and zinc, which slows its absorption and provides a prolonged duration of action compared to regular insulin. Its primary mechanism of action involves binding to the insulin receptor, which facilitates glucose uptake into skeletal muscle and fat while inhibiting hepatic glucose output. It is typically administered subcutaneously and is indicated for glycemic control in both type 1 and type 2 diabetes mellitus.
