Ispinesib is a synthetic small molecule derived from quinazolinone with antineoplastic properties. It acts as a potent, specific, and reversible inhibitor of the mitotic motor protein kinesin spindle protein (KSP), also known as HsEg5. By inhibiting KSP, ispinesib disrupts mitotic spindle assembly, induces cell cycle arrest during the mitotic phase, and leads to cell death in actively dividing tumor cells. Because KSP is not involved in nonmitotic processes such as neuronal transport, ispinesib may have a lower risk of causing peripheral neuropathy compared to tubulin-targeting agents. Ispinesib has been investigated primarily for the treatment of various cancers including breast cancer and renal cell carcinoma[1][3][4][5].
