Overview

ISPM19 is a proprietary **inhaled liposomal hydroxychloroquine** product developed by InspirMed for the prophylaxis and treatment of COVID-19. The drug reformulates hydroxychloroquine in a liposome-based nanomedicine intended for nebulized delivery directly to the airways and lungs, with the goal of achieving local pulmonary exposure while reducing systemic exposure and toxicities associated with oral hydroxychloroquine. Its proposed antiviral mechanism is based on hydroxychloroquine pharmacology, including inhibition of endosomal and lysosomal acidification, interference with intracellular viral entry and uncoating processes, and reduced glycosylation-dependent interaction of the host angiotensin-converting enzyme-2 receptor with coronavirus spike protein. Publicly described development placed ISPM19 in Phase 1 testing in healthy volunteers as an ascending-dose safety, tolerability, and pharmacokinetic study.
