IT848 is a novel, direct inhibitor of the nuclear factor kappa B (NF-κB) signaling pathway, developed by ImmuneTarget Inc. It is specifically designed to block the DNA-binding activity of all five NF-κB subunits (p65, RelB, c-Rel, p50, and p52), thereby suppressing the transcriptional activity of this key oncogenic driver. In addition to its direct inhibitory effects on NF-κB, IT848 disrupts redox homeostasis in multiple myeloma cells by depleting the reduced glutathione pool, leading to selective oxidative stress and apoptosis in malignant cells while sparing healthy ones. Preclinical data in multiple myeloma models demonstrate that IT848 is effective as a single agent and shows synergistic activity when combined with the proteasome inhibitor bortezomib or with PD-1 checkpoint blockade. Correlative immune studies also suggest that IT848 may modulate the tumor microenvironment by suppressing regulatory T-cell infiltration.
