ITC-6102RO is a novel B7-H3-targeted antibody-drug conjugate (ADC) developed for the treatment of solid tumors that express B7-H3, such as breast and lung cancers. It consists of a monoclonal antibody that targets B7-H3 (CD276) conjugated via a highly biostable Ortho Hydroxy-Protected Aryl Sulfate (OHPAS) linker to dHBD—a DNA alkylating agent and derivative of pyrrolobenzodiazepine (PBD). The drug demonstrates high specificity for B7-H3, leading to internalization into tumor cells and delivery of dHBD, causing S phase cell cycle arrest, DNA damage, and apoptosis. Preclinical data show potent antitumor efficacy in B7-H3-positive cell-derived and patient-derived xenograft models with favorable safety profiles[1][3][7][10][12].
