ITC-6146RO is an antibody-drug conjugate (ADC) developed by IntoCell, Inc., targeting B7-H3 (B7 homolog 3), a protein frequently overexpressed in various solid tumors. The drug incorporates IntoCell's proprietary OHPAS (Ortho-Hydroxy Protected Aryl Sulfate) linker system and PMT (Payload Modification Technology) to deliver a highly potent duocarmycin-class DNA-alkylating payload. This design aims to enhance systemic stability and ensure efficient payload release within the tumor environment. ITC-6146RO is currently in Phase 1a/b clinical trials for the treatment of advanced or metastatic solid tumors, including metastatic castration-resistant prostate cancer (mCRPC), non-small cell lung cancer (NSCLC), and triple-negative breast cancer (TNBC), particularly in patients who have failed standard therapies.
