ITF3756 is a selective small molecule inhibitor of histone deacetylase 6 (HDAC6) being developed by Italfarmaco. Unlike pan-HDAC inhibitors, ITF3756 specifically targets the HDAC6 isoform, which plays a key role in regulating cellular processes, gene expression, and chromatin dynamics. Preclinical data suggests that ITF3756 possesses potent immunomodulatory properties; it reduces the immunosuppressive activity of myeloid cells within the tumor microenvironment while simultaneously promoting T cell activation. This dual action makes it a promising candidate for enhancing the efficacy of immune checkpoint blockade therapies. ITF3756 is currently undergoing Phase I clinical evaluation for the treatment of advanced solid tumors.
