ITGA4 ADC is an experimental antibody-drug conjugate (ADC) targeting Integrin alpha-4 (ITGA4, also known as CD49d), currently in preclinical development by Vor Bio for the treatment of acute myeloid leukemia (AML). ITGA4 was identified as a high-priority therapeutic target through a multimodal atlas of AML patient samples, which revealed its consistent expression on leukemic blasts at both diagnosis and relapse. The ADC is designed to selectively bind to ITGA4 on the surface of AML cells and deliver a cytotoxic payload to induce cell death. Preclinical data presented at the ASH 2024 meeting demonstrated that the ITGA4 ADC achieved significant in vitro cytotoxicity against AML cell lines, such as MOLM-13, at sub-nanomolar concentrations. This suggests its potential as a therapeutic agent to address antigen heterogeneity and prevent relapse in AML patients, particularly when used as part of multi-specific immunotherapy strategies.
