ITGB6 ProTriTAC is a preclinical-stage protease-activated trispecific T-cell engager (ProTriTAC) being developed by Harpoon Therapeutics (a subsidiary of Merck & Co.) for the treatment of various solid tumors, including lung, cervical, and head and neck cancers. The molecule is engineered as a prodrug to enhance the therapeutic window of T-cell engagers; it remains systemically inactive until it reaches the tumor microenvironment, where tumor-associated proteases cleave a specific linker to release the active T-cell engaging domains. The construct features three humanized antibody-derived binding domains: one specific for the tumor antigen integrin beta-6 (ITGB6), one for the CD3 epsilon subunit on T cells to trigger activation, and one for serum albumin to extend the drug circulatory half-life. This localized activation strategy is intended to minimize systemic cytokine release and off-tumor toxicities while maintaining potent anti-tumor activity.
