Overview

ITI-009 is a small molecule phosphodiesterase 1 (PDE1) inhibitor developed by Intra-Cellular Therapies. PDE1 enzymes are highly expressed in the brain and play a critical role in regulating signaling pathways involving cyclic nucleotides, specifically cAMP and cGMP. By inhibiting PDE1, ITI-009 aims to enhance intracellular signaling pathways associated with dopamine D1 receptors, which are essential for cognitive and motor functions. The compound has been investigated in preclinical models for the treatment of various central nervous system (CNS) disorders, including Parkinson's disease, schizophrenia, and cognitive impairment. While ITI-009 served as an early representative candidate in Intra-Cellular's PDE1 inhibitor platform, the company has primarily advanced ITI-214 (lenrispodun) as its lead clinical candidate in this class.
