Overview

ITI-1549 is a novel, small molecule, non-hallucinogenic psychedelic under development by Intra-Cellular Therapies for the treatment of mood and other neuropsychiatric disorders. It acts as an agonist at the serotonin 5-HT2A receptor, favoring postsynaptic beta-arrestin pathway signaling over G-protein coupled pathways—a mechanism believed to retain therapeutic benefits of psychedelics without hallucinogenic effects. Additionally, it is a potent agonist at the serotonin 5-HT2C receptor and an antagonist at the serotonin 5-HT2B receptor, which reduces risk for cardiac valvulopathy associated with many serotonergic psychedelics. Preclinical studies show that ITI-1549 increases social interaction and reduces anxiety and depressive symptoms in animal models; it also activates the mTOR pathway in the prefrontal cortex, suggesting potential for rapid antidepressant activity via increased neuroplasticity. As of early 2025, ITI-1549 remains in IND-enabling studies with Phase 1 clinical trials anticipated to begin soon[1][4][5].
