Overview

ITI-2000 is a therapeutic cancer vaccine being developed by Immunomic Therapeutics for the treatment of Human Papillomavirus (HPV)-positive tumors, with a primary focus on head and neck cancers. The vaccine utilizes self-amplifying RNA (saRNA) technology, which enables higher and more sustained levels of antigen expression compared to conventional mRNA. It is engineered using the proprietary UNITE (UNiversal Intracellular Targeted Expression) platform, which involves fusing HPV-derived antigens with the Lysosomal Associated Membrane Protein (LAMP). This fusion directs the expressed antigens into the endosomal/lysosomal compartment of antigen-presenting cells, thereby enhancing MHC Class II presentation. This mechanism is intended to elicit a robust and comprehensive immune response, involving both CD4+ helper T cells and CD8+ cytotoxic T cells, specifically targeting and eliminating HPV-expressing tumor cells.
