ITK-1 is a personalized peptide-based therapeutic cancer vaccine developed to stimulate the immune system against tumor antigens. The vaccine is tailored for each patient by selecting suitable peptides based on their human leukocyte antigen (HLA) type, particularly HLA-A24. Its mechanism of action involves immunostimulation—boosting cytotoxic T lymphocyte responses and inducing peptide-specific immunoglobulin G production. ITK-1 was primarily investigated for use in recurrent or progressive glioblastoma multiforme (GBM) and castration-resistant prostate cancer, especially in patients refractory to standard therapies such as temozolomide or docetaxel. Clinical trials demonstrated that the vaccine was generally safe and well-tolerated, with dose-dependent immune activation observed; however, pivotal phase III studies failed to show statistically significant improvement in overall survival compared to placebo in prostate cancer patients. As a result, development has been discontinued[1][9][10].
