ITM-31 (formerly known as LuCaFab) is a targeted radionuclide therapy (TRT) candidate being developed for the treatment of malignant glioblastoma. It consists of a monoclonal antibody Fab fragment, designated 6A10, which specifically targets carbonic anhydrase XII (CA XII), conjugated to the beta-emitting radioisotope lutetium-177 (n.c.a. 177Lu, EndolucinBeta). CA XII is highly expressed on the surface of glioblastoma cells but is largely absent in healthy brain tissue, allowing for high tumor specificity. The drug is administered via direct intracavitary injection into the tumor resection cavity following standard-of-care treatment. Once administered, ITM-31 binds to residual glioblastoma cells and delivers localized cytotoxic radiation, intended to destroy remaining tumor cells and prevent recurrence. The therapy is the result of a collaboration between ITM Isotope Technologies Munich, Helmholtz Munich, and the University Hospital Münster.
