iTOL-101 is an investigational cell therapy being developed by iTolerance as a potential cure for Type 1 Diabetes. The therapy utilizes allogeneic cadaveric pancreatic islets co-implanted with iTOL-100, an immunomodulatory platform based on a synthetic form of the Fas ligand (FasL) protein combined with a biotin-polyethylene glycol microgel. This combination aims to induce durable, localized immune tolerance at the site of implantation, allowing the transplanted islet cells to function and restore insulin secretion without the need for chronic systemic immunosuppression. Preclinical studies in non-human primates have demonstrated long-term graft function and blood glucose control using this approach.
