Overview

Itruvone is a small molecule investigational drug developed as a nasal spray for the treatment of major depressive disorder. It is classified as a vomeropherine and is administered at microgram-level doses intranasally. The proposed mechanism of action involves binding to receptors on chemosensory neurons in the nasal cavity, which then activate neural circuits in the brain—specifically olfactory-amygdala pathways—believed to increase activity in the limbic-hypothalamic sympathetic nervous system and promote catecholamine release. This mechanism is fundamentally different from currently approved antidepressants, as itruvone does not require systemic uptake or direct brain penetration to exert its effects, potentially reducing side effects associated with other rapid-onset therapies like ketamine-based treatments. Itruvone has completed Phase 2A clinical trials for major depressive disorder and is being advanced into further clinical development by Vistagen[1][4][5][6][8].
