Overview

ITSA-1 is a small molecule that functions as an agonist of Histone Deacetylase 3 (HDAC3). It was originally identified as a chemical tool capable of suppressing the effects of the HDAC inhibitor Trichostatin A (TSA), thereby effectively promoting or restoring HDAC activity. In biomedical research, particularly in respiratory medicine, ITSA-1 is used to investigate the epigenetic regulation of inflammatory pathways. Recent studies have demonstrated that ITSA-1-mediated activation of HDAC3 enhances the deacetylation of IL-33 at lysine 16, which facilitates the release of the cytokine from the nucleus of lung epithelial cells. This mechanism has been shown to exacerbate allergic airway inflammation and increase the severity of asthma in experimental models, identifying the PIM1-HDAC3-IL-33 axis as a potential target for therapeutic intervention.
